As the title says, when taking off my diff seems to knock 3 or 4 times and at this time take off is slow, then after the knocks it is fine for normal driving. It also happens sometimes when under load. Could this be lack of diff oil? or mabye something more sinister?

What diff is in the car, if it's lsd could be broken clutch bolts on the crown wheel and single spinner could be teeth missing off the sun gears.
